This study presents the development of immunoanalytical test system for the detection of undeclared chicken meat additives in food products. It is based on immunochromatographic analysis of immunoglobulins as a biomarker for species identification. The proposed analysis can be applied both for rapid simultaneous authentication of chicken in meat products and for quantitative estimation of its ratio in raw materials. During the assay, immunoglobulins that are contained in muscle tissue and can be easily extracted from meat are detected. A sandwich format of lateral flow assay (LFIA) based on gold nanoparticles as a label and colorimetric detection (both visual and instrumental) is used to determine immunoglobulins and, consequently, a source of meat. It was found that the developed LFIA allows distinguishing poultry (chicken, turkey) meat sources within 15 min. To achieve the highest analytical parameters, the LFIA was optimized by varying combination of immune reactants, test strip composition, and the assay conditions. Different methods for sample preparation of meat products were tested and compared. The developed LFIA was applied to analyze meat foodstuffs and allowed revealing an adulteration with up to 0.1% (w/w) sensitivity thus demonstrating high productivity and accuracy of the results. The proposed test system can be recommended for rapid on-site screening control of the composition and quality of meat products.
